盐酸克伦特罗胶体金快速检测试剂盒质量研究

摘要
选取3种市售同批次盐酸克伦特罗胶体金免疫层析快速检测试剂盒(D、M、A),引入金标读数仪,对其最小检出浓度、灵敏度、特异性、检出限、阳性检出率、假阳性率和假阴性率等方面进行研究,比较3种市售盐酸克伦特罗胶体金免疫层析快速检测试剂盒的质量。结果表明,D试剂盒中的检测卡在检测盐酸克伦特罗标准溶液时产品性能最优,M试剂盒的样品前处理方法和检测卡在猪肉基质中的综合使用效果最优。阴性猪肉加标样本的快速检测结果显示,试剂盒检测结果与金标读数仪器检测结果 100%符合,3种产品的最小检出浓度均小于产品说明书标注的检出限,且灵敏度100%,D、M特异性100%,D、M假阳性率0%,M阳性检出率100%,D、A阳性检出率均大于90%。综合分析,3种试剂盒的准确性、特异性等均较好,阳性检出率、假阴性率均在可接受范围内,其中,M试剂盒产品性能最优,可用于日常生猪屠宰点和农贸市场中猪肉盐酸克伦特罗的初筛。
In this study, three kinds of commercially available clenbuterol hydrochloride colloidal gold immunochromatographic rapid detection kits(D, M, A)was chosed and evaluated in terms of minimum detectable concentration, sensitivity, specificity, the detection limit, positive rate, false positive rate, false negative rate with gel gold mark test strip reading instrument which confirmed the results of the GICA strips. On the quality comparision of the commercially available clenbuterol hydrochloride colloidal gold immunochromatographic rapid detection kits, the optimum performance of the commercially available clenbuterol hydrochloride GICA strip was D detection kit when determination of clenbuterol hydrochloride standard solution, by the way, the optimum performance of the results of pretreatment method and gel gold mark test strip was M detection kit when determination of clenbuterol hydrochloride in pork matrix. The result of the determination of positive pork matrix samples showed that: the accordance rate of the results between the gel gold mark test strip and the gel gold mark test strip reading instrument was 100%,the minimum detectable concentration of the three kind of the rapid detection kits was less than the detection limit indicated in the product description, and the sensitivity was 100%, the specificity of D and M rapid detection kits was 100%,the false positive rate was 100%, the positive rate of M was 100%, the positive rate of D and A was more than 90%. As stated previously, the three detection kits had a good accuracy and specificity,the positive rate and the false negative rate were both in the acceptable. On the quality comparision of three kinds of the rapid detection kits, M detection kit was the best one being used to preliminary test clenbuterol hydrochloride in pork when daily management of pig slaughter sites and farm produce markets.
