摘要
目的探讨槲皮素对糖尿病大鼠肾脏足细胞nephrin和podocin表达的影响。方法采用腹腔注射链脲佐菌素(STZ)方法构建糖尿病大鼠模型,分为正常对照组、糖尿病模型组、槲皮素给药组。12周后测定肾脏指数、血肌酐、尿素水平及24h尿中总蛋白、白蛋白、肌酐、尿素水平;Western blot方法检测肾皮质nephrin和podocin的蛋白表达。结果槲皮素给药组肾脏指数、血肌酐和尿素水平低于糖尿病模型组;槲皮素给药组尿中总蛋白、白蛋白水平明显低于糖尿病组,尿中肌酐、尿素水平高于糖尿病组。槲皮素给药组肾皮质nephrin和podocin的蛋白表达水平明显高于糖尿病模型组。结论槲皮素对糖尿病大鼠肾脏功能具有保护作用,槲皮素可能是通过增强足细胞nephrin和podocin表达发挥肾脏保护作用。
        Objective To study the effect of quercetin on the expression of nephrin and podocin in renal podocytes of diabetic rats.Methods The diabetic rat model was established by intraperitoneal injection of streptozotocin(STZ),which was divided into normal control group,diabetic group and quercetin group.After 12 weeks,kidney weight/body weight,serum creatinine,urea levels and 24-hour urine total protein,albumin,creatinine and urea levels were measured.The protein expression of nephrin and podocin in renal cortex was detected by Western blot.Results The levels of kidney weight/body weight,serum creatinine and urea in quercetin group were lower than those in diabetic group;the levels of total protein and albumin in urine in quercetin group were lower than those in diabetic group,and the levels of creatinine and urea in urine were higher than those in diabetic group.The protein expression levels of nephrin and podocin in renal cortex of quercetin group were significantly higher than those of diabetic group.Conclusion Quercetin has a protective effect on renal function in diabetic rats.Quercetin may play aprotective role by enhancing the expression of nephrin and podocin in podocytes.
