摘要
为了给苔藓结皮野外恢复的大规模接种提供丰富种源,本文利用0.1%NaClO(10、15、20、30、45、60、120s)和0.1%HgCl_2(10、15、20、30、45、60、120 s)两种消毒液,采用Knop、MS和Hoagland 3种培养基,设置5.5、6、6.5、7、7.5、8、8.5七个水平的p H值,通过单因素试验设计方法,测定长尖对齿藓[Didymodon ditrichoides(Broth.)]茎叶体的成活率、原丝体长度和分枝数等指标,探讨影响长尖对齿藓原丝体扩繁的关键因子。结果表明:(1)消毒方式、培养基及p H均对长尖对齿藓茎叶体的成活率、原丝体长度和分枝数有显著影响(P<0.05),影响大小依次均为消毒方式>培养基>p H;(2)最适合的消毒方式为:0.1%NaClO消毒15~20 s;(3)最适合长尖对齿藓原丝体生长的培养基是Knop和Hoagland培养基;(4)最适合长尖对齿藓原丝体生长的pH是7.5。从而得出快速培育长尖对齿藓原丝体的最佳组合为:0.1%NaCl O消毒15~20 s+Hoagland/Knop+pH=7.5。本文的研究结果可以缩短长尖对齿藓生长周期以进行快速繁殖,为苔藓结皮的快速培育以及工程化应用提供一定的借鉴。
        In order to provide a rich source of mass vaccination for the field restoration of moss crust,we used two kinds of disinfection method,0. 1% NaClO(10,15,20,30,45,60,120 s) and 0. 1% HgCl_2(10,15,20,30,45,60,120 s),with Knop,MS and Hoagland as culture media,and set the p H values(5. 5,6,6. 5,7,7. 5,8,8. 5),through the single factor experiment design method,we determined the survival rate of the cormus of [Didymodon ditrichoides( Broth.)],protonema length and number of branches,thereby exploring key factors that affect the propagation of the protonema of D. ditrichoides. The results are as follows:(1) The disinfection method,culture medium and p H value have significant influence upon the survival rate of the cormus of D. ditrichoides,length of protonema and number of branches( P < 0. 05),in a descending order of disinfection method,culture medium,and pH value;(2) The most suitable disinfection method is 0. 1% NaClO with a duration of 15-20 s;( 3) The most suitable culture medium for the growth of the protonema of D.ditrichoides is Knop and Hoagland;(4) The most suitable p H value for the growth of protonema of D. ditrichoidesis 7. 5. Therefore,the best combination of factors for rapid cultivation of the protonema of D. ditrichoides is0. 1% NaClO disinfection 15-20 s + Hoagland/Knop + pH = 7. 5. Our experiment can be determined to shorten the growth cycle of D. ditrichoides and realize rapid propagation,thereby providing experience for the rapid culture and engineering application of moss crust.
