摘要
目的依据"肾主骨、生髓、通于脑"的理论,通过去卵巢方式建立骨质疏松症大鼠动物模型,探讨该病证动物模型神经内分泌方面的病理机制。方法创建正常组(A组)、模型组(B组)、雌激素组(C组)及胎肾组(D组)。应用胎肾细胞悬液治疗去卵巢骨质疏松大鼠(D组)。采用放免法测定E2、GH水平,免疫组化法测定DAR、AR、5-HTR的表达,并进行组间比较。结果 B组的E2、GH水平均低于A组,C组和D组的E2、GH水平均高于B组(P<0.05)。B组的DAR、AR、5-HTR表达高于A组,C组和D组的DAR、AR、5-HTR表达低于B组(P<0.05)。结论骨质疏松症的病理机制与神经内分泌网络具有密切的相关性,胎肾具有治疗大鼠去卵巢所致骨质疏松的作用,可能对神经系统的调控机制也发挥一定的作用。
        Objective To establish animal models of osteoporosis by ovariectomy according to the theory "kidney determining the condition of the bone and marrow, and connects to brain", and to explore the neuroendocrine pathological mechanism of the animal model of osteoporosis. Methods The normal group(group A), model group(group B), estrogen group(group C) and fetal kidney group(group D) were established. Fetal kidney cell suspension was used to treat ovariectomized osteoporotic rats(the group D). The levels of E2 and GH were measured by radioimmunoassay, the expressions of DAR, AR and 5-HTR were determined by immunohistochemistry, and these indexes were compared among the groups. Results The levels of E2 and GH in the group B were lower than those in the group A, while those in the groups C and D were higher than the group B(P<0.05). The expressions of DAR, AR and 5-HTR in the group B were higher than those in the group A, while those in the groups C and D were lower than the group B(P <0.05). Conclusion The pathological mechanism of osteoporosis is closely related to neuroendocrine network. Fetal kidney can treat ovariectomized osteoporosis rats, and may also play a role in regulating the nervous system.
