HPLC法同时测定康溃丸中黄芪甲苷藁本内酯和党参炔苷的含量

摘要
目的建立HPLC法同时测定康溃丸中黄芪甲苷、藁本内酯、党参炔苷的含量的方法。方法采用高效液相色谱法,色谱条件:Agilent Zorbax Eclipse XDB-C_(18)色谱柱(4.6 mm×250 mm,5μm);混合流动相:流动相A为乙腈,流动相B为0.25 moL·L~(-1)三氟乙酸水溶液,梯度洗脱;流速:1.0 mL·min~(-1);柱温:28℃;进样量:10μL;检测波长:282nm。结果黄芪甲苷、藁本内酯、党参炔苷检测质量浓度线性范围分别为2.256～32.158 μg·mL~(-1),2.268～58.142 μg·mL~(-1),2.284～46.524 μg·mL~(-1)。平均加样回收率(RSD)分别为100.95(0.88)、99.42(1.06)、99.42(0.92)。结论该方法简单快速、准确可靠、专属性、精密度、稳定性、重复性均较好,为康溃丸质量标准的提高提供有意义的实验基础。
Objective:To establish a HPLC method for simultaneous determination of the contents of astragaloside,ligustilide and codonopsis in Kangchuan Pills.Methods:HPLC was used with chromatographic conditions,Agilent Zorbax Eclipse XDB-C18 column(4.6 mm×250 mm,5 μm).Mixed mobile phase:mobile phase A was acetonitrile and mobile phase B was 0.25 moL·L~(-1) aqueous solution of trifluoroacetic acid.gradient elution,1.0 mL·min~(-1) of flow rate,28°C of column temperature,10 μL of injection amount and 282 nm of detection wavelength.Results:The linear range of the detection quality concentration of astragaloside,ligustilide and ginsenoside were from 2.256 to 32.158 μg·mL~(-1),2.268 to 58.142 μg·mL~(-1) and 2.28 to 46.524 μg·mL~(-1),respectively.The average sample recovery(RSD)was 100.95(0.88),99.42(1.06),and 99.42(0.92),respectively.Conclusion:The method is simple,rapid,accurate and reliable,and has good specificity,precision,stability and repeatability and provides a meaningful experimental basis for the improvement of Kangkui Pill quality standard.
