摘要
为了探讨鸡Toll样受体3(chicken toll-like receptor 3,chTLR3)在传染性法氏囊病病毒(infectious bursal dis-ease virus,IBDV)感染中的作用及microRNA(miRNA)对其调控的分子机制,本研究用IBDV感染DF-1细胞,qRT-PCR和Western blot分析表明IBDV感染可使DF-1细胞中chTLR3的表达水平显著升高。chTLR3激活表达后可抑制IBDV的复制,进一步分析其机理发现,chTLR3表达上调可激活IFN-β启动子和抗病毒天然免疫基因Mx启动子的活性及NF-κB和IRF-7信号通路。用生物信息学方法和双荧光素酶报告系统分析发现gga-miR-6655-5p可作用于chTLR3的3′UTR;qRT-PCR和Western blot分析表明gga-miR-6655-5p可以使chTLR3的蛋白水平表达降低,但mRNA水平没有变化。本研究表明:gga-miR-6655-5p可以作用于chTLR3的3′UTR负调控chTLR3的表达,抑制IFN-β启动子和Mx启动子活性,使IBDV在细胞中的复制水平提高。
        The innate immune system acts as the first line of defense against invading viruses and plays an important role in the subsequent activation of antiviral responses.Chicken toll-like receptor 3(chTLR3)is a critical pattern recognition receptors(PRRs)to sense the pathogen-associated molecular patterns(PAMPs)of RNA virus and initial an antiviral innate immune response in chicken.microRNAs(miRNA)are involving in the regulation of chTLR3 to recognize RNA virus infection.However,its role of antiviral activity against infectious bursal disease virus(IBDV)infection and how miRNA regulate chTLR3 in chicken cells both have not been clarified.In this study,we found that the expression of chTLR3 in IBDV-infected-DF-1 cells was significantly increased.Activated chTLR3 signaling via chTLR3 ligand stimulaiton could activate IFN-βpromoter and Mx promoter through IRF7-and NF-κB-dependent pathway and inhibit the replication of IBDV in DF-1 cells,whereas suppression of chTLR3 by siRNA could lead to the contrary results.In addition,we proved that gga-miR-6655-5 p can posttranscriptionally regulate the chTLR3 protein expression level through inhibiting the translation of chTLR3 mRNA based on dual-luciferase reporter system,qRT-PCR and Western blot analysis.Overexpression gga-miR-6655-5 p,reduced the expression of chTLR3 protein,which promoted IBDV replication and decreased the activity of IFN-βpromoter and Mx promoter.In conclusion,Our results suggested that gga-miR-6655-5 p is a negative regulator of chTLR3 and promotes IBDV replication in DF-1 cells.
