摘要
目的探讨间歇性低压低氧预处理对SD大鼠放射性肝损伤的保护作用。方法将24只雄性SD大鼠随机分为对照组(C组)、4 000m低压低氧预处理组(IHHP组)、单纯照射组(R组)和低压低氧预处理联合照射组(IHHP+R组),每组6只,对照组大鼠在常规环境下饲养,IHHP组大鼠通过低压低氧舱模拟海拔高度4 000m的高原环境,6h/次/d,5次/w,照射组大鼠照射总剂量DT=60Gy,2Gy/次/d,5次/w,照射完成后6h采下腔静脉血检测谷丙转氨酶(ALT)、谷草转氨酶(AST),肉眼观察大鼠肝脏的变化,HE染色显微镜下观察肝组织的结构变化。结果 IHHP+R组与R组比较,2组间ALT、AST值降低,差异有统计学意义(P<0.05)。肉眼可见IHHP+R组大鼠肝脏轻度充血,HE染色IHHP+R组肝脏中央静脉、肝窦扩张充血,肝细胞空泡样变性数量及程度较R组明显减轻。结论间歇性低压低氧预处理后对放射造成的SD大鼠的肝损伤有保护作用。
        Objective To investigate the protective effect of intermittent hypobaric hypoxia preconditioning on radiation induced liver injury in SD rats.Methods 24 male SD rats were randomly divided into control group(C),hypobaric hypoxia preconditioning group(IHHP),irradiation group(R)and hypobaric hypoxia preconditioning combined with irradiation group(IHHP+R),and 6 rats in each group.The rats in C group were housed under normal conditions.The rats in IHHP group were challenged by low pressure hypoxia chamber simulated altitude of 4 000 maltitude,6 hours/time/day,5 times/week.The irradiation group rats irradiated with a total dose of DT=60 Gy,2 Gy/f/d,5 f/w.Six hours after the irradiation,venous blood was collected for detection of alanine aminotransferase(ALT)and aspartate aminotransferase(AST),and the changes of the liver of the rats were observed.The changes of liver tissues were observed by HE staining.Results IHHP+R group compared with the R group,the ALT and AST values of the two groups decreased,and the difference was statistically significant(P <0.05).In the IHHP+R group,the liver was slightly congested,and HE staining showed that the number and degree of hepatocyte vacuolar degeneration in group IHHP+R were significantly lower than those in R group.Conclusion Intermittent hypobaric hypoxia preconditioning has protective effects on radiation induced liver injury in SD rats.
