摘要
目的:探讨滋阴养血安神方对睡眠剥夺小鼠睡眠的影响及其机制。方法:利用腹腔注射对氯苯丙氨酸(PCPA)制造睡眠剥夺模型,模定失眠小鼠戊巴比妥钠致睡眠的阈下及阈上剂量,分6组,空白对照组、模型组、滋阴养血安神方低剂量组(73 g/kg)、滋阴养血安神方中剂量组(146 g/kg)、滋阴养血安神方高剂量组(292 g/kg)、地西泮组(1.5 mg/kg),造模并给药7 d后,给予戊巴比妥钠诱导睡眠,观察对小鼠睡眠的影响,并检测小鼠海马5-羟色胺(5-HT)含量。结果:滋阴养血安神方可增加阈下剂量戊巴比妥钠小鼠睡眠个数,缩短阈上剂量戊巴比妥钠的小鼠入睡潜伏期,与模型组对比有显著性差异(P<0.01),能延长阈上剂量戊巴比妥钠诱导的睡眠时间,与模型组对比有显著性差异(P<0.01);滋阴养血安神方高剂量组小鼠脑内5-HT含量显著高于模型组,与模型组对比有显著性差异(P<0.05)。结论:滋阴养血安神方具有镇静、催眠作用,对失眠症的有一定的治疗作用,其对小鼠睡眠的影响机制之一可能是通过升高脑内海马5-HT含量实现的。
        Objective:To explore the effect of ZiyinYangxueAnshen Decoction on sleep deprivation in mice and its mechanism.Method:The model was established by receiving PCPA, the subthreshold and upper threshold dose of pentobarbital sodium induced sleep in the model insomnia mice were decided.Then six groups such as blank control group, model group, the low dose group of ZiyinYangxueAnshen Decoction(73 g/kg),the dose group of ZiyinYangxueAnshen Decoction(146 g/kg), the high dose group of ZiyinYangxueAnshen Decoction(292 g/kg)and diazepamgroup(1.5 mg/kg) were established. After each drug administration group was given 7 days, pentobarbital sodium was given to induce sleep. Then we observed the sleeping function effects on sleep deprivation mice.The contents of 5-hydroxytryptamine(5-HT) in mice's hippocampus were detected. Result:ZiyinYangxueAnshen Decoction can increase the number of subthreshold dose of sodium pentobarbital mice sleep and shorten the threshold dose of pentobarbital sodium on mice sleep latency, which had significant difference from the model group(P<0.01).ZiyinYangxueAnshen Decoction can also prolong the sleeping timeinduced by pentobarbital sodium compare with the model group(P<0.01).The content of 5-HT detected by ELISA in the high dose of ZiyinYangxueAnshen Decoction was significantly higher than that of model group(P<0.05).Conclusion:ZiyinYangxueAnshen Decoction has sedative and hypnotic effects, and has certain therapeutic effects on insomnia.One of the possible mechanisms that may influence sleep in mice is by increasing the level of 5-HT in the hippocampus.
