除幽愈疡免煎颗粒对胃溃疡模型大鼠的实验研究

摘要
目的研究除幽愈疡免煎颗粒对乙酸致大鼠实验性胃溃疡的愈合效果。方法将50只SD大鼠按随机数字表法分为5组:正常组、模型组、西药组、中西医结合组及中药组,每组10只,采用冰乙酸腐蚀法制作胃溃疡大鼠模型。各组大鼠于术后第2天,西药组予奥美拉唑0. 132 mg/100 g灌胃,中西医结合组按照西药组基础上加用除幽愈疡免煎颗粒,0. 5 mL/100 g灌胃,中药组予0. 5 mL/100 g除幽愈疡免煎颗粒灌胃,正常组和模型组予1. 25 mL/100 g生理盐水灌胃,每天1次,连续干预14天。观察各组大鼠胃黏膜病理形态学变化;计算溃疡面积和溃疡抑制率; ELISA方法检测血清标本碱性成纤维细胞生长因子(bFGF)含量变化;免疫组化法检测各组大鼠bFGF蛋白表达,Real-time PCR法检测各组大鼠bFGF mRNA表达。结果与正常组比较,模型组大鼠胃壁结构破坏,黏膜出现坏死、缺损,腺体消失,细胞排列紊乱,黏膜层有炎性细胞浸润;模型组bFGF蛋白表达较弱,血清bFGF含量和bF-GF mRNA表达水平均降低(P <0. 05)。与模型组比较,光镜下各药物干预组胃壁结构和黏膜恢复较好,溃疡面积均缩小(P <0. 01,P <0. 05),中药组较西药组和中西医结合组溃疡面积减小,抑制率明显升高(P <0. 05)。与模型组比较,中药组和中西医结合组大鼠胃黏膜组织bFGF蛋白表达水平较强;中西医结合组血清bFGF水平升高,且高于中药组和西药组(P <0. 05);中药组和中西医结合组bFGFmRNA表达升高,且均高于西药组(P <0. 05)。结论除幽愈疡免煎颗粒可增强胃溃疡大鼠胃组织bFGF蛋白及bFGF mRNA表达,调节细胞生长和分化,促进溃疡愈合。
        Objective To study the healing effect of Chuyou Yuyang Granule( CYG) for the experimental gastric ulcer induced by acetic acid in rats. Methods Totally 50 SD rats were randomly divided into 5 groups,i. e.,the normal group,the model group,the Western medicine treatment group,the integrated Chinese and Western medicine group,and the Chinese medicine treatment group,10 in each group. The rat model of gastric ulcer was established by means of glacial acetic acid corrosion method. The rats in each group were performed intragastric administration at the second day after the operation,Rats in the Western medicine group was administered omeprazole 0. 132 mg/100 g by gastrogavage. In accordance with the intervention of Western medicine group,the integrated Chinese and Western medicine group added the CYG,and 0. 5 m L/100 g was given for intragastric administration. The Chinese medicine group was given 0. 5 m L/100 g CYG for intragastric administration,the normal group and the model model were given 1. 25 m L/100 g physiological saline. The intervention was performed once a day for 14 consecutive days. The pathological changes of gastric mucosa in the ras of each group were observed,ulcer area and the ulcer inhibition rate of each treatment group was calculated. The changes of bFGF content in the serum samples were measured by ELISA method; the expression of b FGF protein in each group was detected by immunohistochemical method,and the expression of b FGF mRNA in each group was detected by Real-time PCR. Results Compared with the normal group,the gastric wall structure of the model group was destroyed,the mucous membrane was necrotic and defective,the gland disappeared,the cells were disarranged and the mucous layer was infiltrated by inflammatory cells; the expression of b FGF protein in the model group was relatively weak; the content of serum bFGF of the model rats decreased significantly(P < 0. 05),and the expression level of b FGF mRNA decreased(P < 0. 05). Compared with the model group,the gastric wall structure and mucous membrane recovery were better and the area of the ulcer of various drug intervention group was reduced(P < 0. 01,P < 0. 05) under the light microscope. Compared with the Western medicine group and the integrated traditional Chinese and Western medicine group,the inhibition rate of the Chinese medicine group was significantly increased(P < 0. 05). Compared with the model group,the expression level of bFGF protein in the gastric mucosa of the Chinese medicine group and the integrated Chinese and Western medicine group was stronger; The level of serum bFGF was higher in the integrated Chinese and Western medicine group,and was higher than that of the Chinese medicine group and the Western medicine group(P < 0. 05); the expression of bFGF mRNA in the Chinese medicine group and the Western medicine group increased,and was higher than that in the Western medicine group(P < 0. 05). Conclusion CYG could enhance the expression of bFGF protein and bFGF mRNA in gastric tissue of rats with gastric ulcer,regulate the growth of cell and differentiation,and promote the healing of ulcer.
