摘要
目的探讨肿瘤坏死因子α-238/-308(TNF-α-238/308)基因位点多态性与乳腺癌患者易感性及临床病理特征的相关性,以此为临床上乳腺癌基因筛查提供参考依据。方法选择中国福建泉州地区134例乳腺癌患者及150例健康体检者为研究对象,采用DNA测序方法检测TNF-α启动子基因G238A、G308A位点的基因多态性,应用统计学方法分析基因型分布于乳腺癌发病风险及临床病理特征的关系。结果 TNF-α-238/308基因型GG、GA、AA的频率在2组中差异无统计学意义(P>0.05)。TNF-α-238/308基因多态性与Cerb B-2、肿瘤分期、肿瘤大小、淋巴结转移及雌孕激素受体状态等参数均无相关性(P>0.05)。结论乳腺癌易感性与TNF-α-238/308基因多态性无相关性,因此不推荐将其单独作为乳腺癌基因筛查的候选指标。
        Objective To explore the associations of clinicopathological features and TNF- α- 238 /308 gene polymorphisms with breast cancer susceptibility,so as to provide reference for gene screening of clinical breast cancer. Methods DNA Sequencing technique was used to detect the gene polymorphism of TNF- α- 238 /308 in 134 cancer women with breast cancer and 150 healthy women in Quanzhou,Fujian of China,and statistical analysis was employed to analyze the associations of genotype on the risk of breast cancer with clinicopathological features. Results The frequency differences of TNF- α- 238 or-308 polymorphism genetypes( GG,GA,AA) were not statistically significant between cases group and the control group( P >0. 05). TNF- α- 238 /308 had no correlation with estrogen receptor status,progesterone receptor status,Cerb B- 2 status,lymphnode metastases,tumor size or tumor stage( P > 0. 05). Conclusion TNF- α- 238 /308 polymorphisms has no associations with breast cancer susceptibility,so,it is not recommended to be used as a candidate marker for breast cancer screening.
