摘要
目的目前关于TNF-α基因238位点多态性与女性乳腺癌易感性的报道不一致。本研究通过Meta分析的方法探讨TNF-α基因238位点多态性与乳腺癌易感性的关系。方法通过检索万方、维普、PubMed和Medline数据库,检索时间为1996-01—2016-12,获取相关文献及研究结果,以乳腺癌组与对照组人群TNF-α基因SNP-238位点基因型分布的OR值为效应指标,采用Stata 11软件进行荟萃分析。结果 Meta分析共纳入9篇文献,累计共77 181例,包括37 429例病例和39 752例对照。等位基因G与A比,OR值为0.980,95%CI:0.960~1.000,P<0.001。亚组分析中亚洲人G等位基因相对于A等位基因OR值为0.982,95%CI:0.782~1.233(P<0.001),而欧美洲人对应的结果 OR值为0.979,95%CI:0.958~1.002,P=0.202。结论总体上,TNF-α基因238位点多态性与乳腺癌易感性无关,不推荐作为筛选乳腺癌基因的候选指标。
OBJECTIVE At present,there is no consistent report on the polymorphisms of the TNF-α-238 and the susceptibility of female breast cancer.This study was to study the relation of genetic polymorphism of TNF-α-238 and breast cancer risk.METHODS All the published studies about TNF-α-238 and breast cancer for the last 20 years were reviewed and standard techniques of meta-analysis to combined with the results of these studies were used to produce a more precise estimate of the significant expression of TNF-α-238.Meta-analysis odds ratios(ORs)were estimated using a fixed effects model or random effects model.RESULTS Nine eligible reports were identified,concerning about 37 429 cancer cases and 39 752 controls.The pooled OR of G allele compared with that of A allele was 0.980(95%CI:0.960-1.000,P<0.001).The pooled OR of G allele compared with that of A allele for Caucasian was 0.979(95%CI:0.958-1.002,P=0.202),for Asian was 0.982(95%CI:0.782-1.233,P<0.001).CONCLUSIONS This meta-analysis suggests that the TNF-a-238 Gallele may not be significantly associated with breast cancer.TNF-a-238 gene is not recommended as a candidate for screening breast cancer genes.Further studies are needed to explore gene-gene and gene-environment interactions.
