摘要
目的观察黄葵总黄酮对慢性肾小球肾炎大鼠的疗效,并对其作用机制进行分析。方法选取50只SD大鼠,随机分为对照组、模型组以及低、中、高剂量组,每组10只。采用小牛血清白蛋白注射法建立慢性肾小球肾炎大鼠模型。低、中、高剂量组分别给予50 mg/kg、100 mg/kg和200 mg/kg黄葵总黄酮灌胃,对照组与模型组给予等体积生理盐水灌胃。测定各组24 h尿蛋白定量、血肌酐、尿素氮、肿瘤坏死因子-α(TNF-α)、白介素-6(IL-6)和T淋巴细胞亚群水平;并检测大鼠肾脏组织TNF-α和IL-6 mRNA水平。结果与对照组比较,模型组的24 h尿蛋白定量、尿素氮、血肌酐、TNF-α、IL-6、CD8+以及肾脏组织TNF-α和IL-6 mRNA水平升高,外周血CD3+、CD4+和CD4+/CD8+比值下降(P<0.05)。与模型组比较,低、中、高剂量组的24 h尿蛋白定量、尿素氮、血肌酐、TNF-α、IL-6、CD8+以及肾脏组织TNF-α和IL-6 mRNA水平下降,外周血CD3+、CD4+和CD4+/CD8+比值显著升高(P<0.05),且低、中、高剂量组上述指标比较差异均有统计学意义(P<0.05)。结论黄葵总黄酮能够有效调节慢性肾小球肾炎大鼠各项生化指标和免疫功能,抑制炎性反应。
        Objective To observe effect of Abelmosehus Moschatus Total Flavonoids in treatment of rats with chronic glomerulonephritis and to analyze its mechanism. Methods A total of 50 SD rats were randomly divided into control group, model group, low, medium and high dose groups(n=10 for each group). Rat models of chronic glomerulonephritis were established by injecting calf serum albumin. Low, medium and high dose groups were lavaged with 50 mg/kg, 100 mg/kg and 200 mg/kg Abelmosehus Moschatus Total Flavonoids respectively, while control and model groups were lavaged with same volumes of physiological saline. Levels of 24-h urinary protein quantitation, serum creatinine, urea nitrogen, tumor necrosis factor-α(TNF-α), interleukin-6(IL-6) and T lymphocyte subsets were detected, and levels of TNF-α and IL-6 mRNA in kidney tissues of rats were also detected in each group. Results Compared with those in control group, levels of 24-h urinary protein quantification, urea nitrogen, serum creatinine, TNF-α, IL-6 and CD8+, and TNF-α and IL-6 mRNA levels in kidney tissues were significantly increased, while values of CD3+, CD4+ and CD4+/CD8+ ratio in peripheral blood were significantly decreased in model group(P<0.05). Compared with those in model group, levels of 24-h urinary protein quantification, urea nitrogen, serum creatinine, TNF-α, IL-6 and CD8+ among low, medium and high dose groups, and TNF-α and IL-6 mRNA levels in kidney tissues were significantly decreased, while values of CD3+, CD4+ and CD4+/CD8+ ratio in peripheral blood were significantly increased(P<0.05), and there were significant differences in the above indexes among low, medium and high dose groups(P<0.05). Conclusion Abelmosehus Moschatus Total Flavonoids may effectively regulate biochemical index and immune function of rats with chronic glomerulonephritis and inhibit inflammatory reaction.
