右归丸药物血清对高糖缺氧条件下大鼠视网膜微血管内皮细胞增殖及迁移的影响

摘要
目的:观察右归丸药物血清对高糖缺氧条件下SD大鼠视网膜微血管内皮细胞增殖及迁移的影响。方法:采用33mmol/L的葡萄糖和100μmol/L的CoCl2同时附加于培养基中培养细胞,以建立体外高糖缺氧的细胞模型,以此为研究对象,分别采用四甲基偶氮唑蓝(MTT)法和Transwell小室法观察不同剂量的右归丸药物血清对模型细胞增殖和迁移的影响。结果:第1天时,右归丸药物血清中剂量组对高糖缺氧抑制细胞增殖起保护作用(P<0.05);第2天时,空白对照组、模型组、右归丸药物血清各剂量组对细胞的增殖作用差异无统计学意义;第4天时,右归丸药物血清高、中剂量组对高糖缺氧引起的细胞增殖起抑制作用(P<0.05)。第2天时,右归丸药物血清中、高剂量组均能降低高糖缺氧对细胞迁移的抑制作用(P<0.003);第4天时,右归丸药物血清低、中、高3个剂量组均能够抑制高糖缺氧对细胞迁移的促进作用(P<0.003)。结论:右归丸药物血清可抑制高糖缺氧条件下SD大鼠视网膜微血管内皮细胞增殖及迁移,减少视网膜新生血管的生成,为右归丸治疗糖尿病视网膜病变提供了科学依据。
        Objective: To observe the effect of Yougui Pills drug serum on proliferation and migration in SD rat retinal microvascular endothelial cells under high glucose and hypoxia condition. Methods: The high glucose and hypoxia model of cell in vitro was established with 33 mmol/L glucose and 100μmol/L CoCl2 simultaneously cultured, and it was used as a research object, by using MTT and Transwell method to obsreve theeffects of different dosage of Yougui Pills drug serum on proliferation and migration in model cells. Results: On the first day, the middle-dose group of Yougui Pills have a protective effect on cell proliferation induced by high glucose and hypoxia(P<0.05); on the 2 nd day, there was no difference in the proliferation of the cells between the blank group, model group, and the each dose group of medicine serum; On the 4 th day, the middle and high dose group medical serum of Yougui Pills inhibited cell proliferation induced by high glucose hypoxia(P<0.05); on the 2 nd day,the middle and high dose group medical serum of Yougui Pill could reduce the inhibitory effect of high glucose and hypoxia on cell migration(P<0.003); On the 4 th day, the three doses of low, medium and high serum of Yougui Pills could inhibit the promotion of cell migration by high glucose and hypoxia(P<0.003). Conclusion: The Yougui Pills medicine serum can inhibit the proliferation and migration of retinal microvascular endothelial cells in SD rats under high glucose and hypoxia conditions, and reduce the production of retinal neovascularization, which provides the scientific basis for treatment of diabetic retinopathy.
