摘要
目的:研究葡萄籽提取物(GSE)对鼻咽癌(NPC)细胞HIF-1α和VEGF表达的影响,为GSE在NPC治疗中的应用提供依据。方法:采用100μmol/L CoCl_2建立细胞拟缺氧模型,用终浓度为0mg/L、12.5 mg/L、25 mg/L和50 mg/L的GSE加入鼻咽癌CNE-2Z细胞系,作用48h后,采用实时荧光定量PCR(Quantitative Real-time PCR)检测低氧诱导因子-1α(HIF-1α)和血管内皮生长因子(VEGF)m RNA表达,免疫印迹技术(Western Blot)检测HIF-1α和VEGF表达。结果:低氧环境下,不同浓度GSE干预抑制鼻咽癌细胞HIF-1α和VEGF表达均受到显著影响。结论:GSE对鼻咽癌CNE-2Z细胞HIF-1α和VEGF表达具有调节作用。
        Objective:To investigate the effect ofgrape seed extract(GSE)on expression of HIF-1α and VEGF gene in nasopharyngeal carcinoma cell line CNE-2Z. Methods:The model of hypoxia was established by 100μmol/L CoCl_2,The GSE with different final concentration of 0mg/L,12.5mg/L,25mg/L and 50mg/L was added to CNE-2Z cells cultured in vitro for succedent experiments.The experimental and blank control groups of CNE-2Z cells were cultivated for 48 h.The expression of HIF-1α and VEGF gene was detected by Quantitative Real-time PCR and Western Blot at m RNA and protein level. Results:Under hypoxic conditions,different solubility of GSE intervention on nasopharyngeal carcinoma cells HIF-1α and VEGF expression were significantly affected. Conclusion:GSE has a regulatory effect on the expression of HIF-1α and VEGF in CNE-2Z cells of nasopharyngeal carcinoma.
