摘要
目的研究AB-8大孔树脂纯化金樱子黄酮的工艺。方法采用总黄酮和槲皮素的质量分数为指标,筛选金樱子的纯化工艺;采用HPLC指纹图谱方法,研究金樱子纯化过程中各样品黄酮类成分特征峰的保留率。结果金樱子纯化最佳工艺条件为:上样液的质量浓度为0.30 g/mL,吸附流速为1.0 mL/min,每1 mL湿树脂的负载量为1.25 mL(相当于0.375 g生药),首先用120 mL水洗脱,再用160 mL的体积分数70%乙醇洗脱。总黄酮保留率为75.33%,槲皮素的保留率为91.94%,黄酮类成分特征峰1~4的保留率超过90%,纯化后总黄酮和槲皮素的精制率分别为2.54倍和3.34倍。结论 AB-8型大孔吸附树脂可以较好地纯化金樱子总黄酮。
        Objective To study the technologic conditions of purifying the total flavonoids from Rosa Laevigata by AB-8 macroporous adsorption resin. Methods The contents of total flavonoids and quercetin were used to screen the purification process of Rosa Laevigata,and the retention rate of characteristic peaks of flavonoids in each sample during the purifying process was studied by the fingerprint of HPLC. Results The optimum technological conditions for the purification of Rosa Laevigata were as follows: the sample concentration was 0.30 g/mL,the adsorption rate was 1.0 mL/min,the load of wet resin per mL was 1.25 mL( equivalent to 0.375 g Rosa Laevigata). First,it was eluted with 120 mL water,and then eluted with 160 mL 70% ethanol. The retention rates of total flavonoids and quercetin were 75.33% and 91.94%. The retention rates of characteristic peaks 1-4 were more than 90%. The refining rates of total flavonoids and quercetin were 2. 54 and 3. 34 times. Conclusion AB-8 macroporous resin can be used to purify total flavonoids of Rosa Laevigata.
