摘要
目的:优化辣蓼黄酮的分离提取工艺以及定量分析其中3种黄酮类组分。方法:采用水煎煮、醇提、醇提后有机溶剂萃取、醇提及有机溶剂萃取后大孔树脂吸附共4种提取方法分离提取辣蓼黄酮,以紫外分光光度法测定总黄酮含量,通过高效液相色谱法测定辣蓼黄酮中3种黄酮类组分(芦丁、金丝桃苷和槲皮苷)的含量。结果:上述4种分离提取方法所得辣蓼总黄酮含量在1.33%~4.92%,所得总黄酮提取物中芦丁含量在0.1155~0.3648 mg/mL,金丝桃苷含量在0.0055~0.0140 mg/mL,槲皮苷含量在0.0141~0.6613 mg/mL。结论:不同提取工艺对总黄酮得率不同,对3种黄酮类组分的提取效果也有差异。比较试验中所用4种方法,醇提法对辣蓼总黄酮以及辣蓼黄酮中的芦丁、金丝桃苷分离提取效果最理想,而对槲皮苷分离提取效果最优的方法则为水煎煮法。
        Objective:To optimize the separation and extraction process of total flavonoids from Polygonum hydropiper L. and quantitative analysis of three flavonoids. Methods:Separation and extraction of PoLygonum hydropiper L. by water decoction,alcohol extraction,organic solvent extraction after alcohol extraction,extraction with macroporous resin after extraction with alcohol mentioned organic solvent. The content of total flavonoids was determined by ultraviolet spectrophotometry. The content of three flavonoids(rutin,hyperoside and quercetin)was determined by HPLC. Results:The content of total flavonoids of PoLygonum hydropiper L. was 1.33%-4.92% in above four extraction methods. The content range of rutin was 0.1155-0.3648 mg/mL,the content range of hyperoside was 0.0055-0.0140 mg/mL,and the content range of quercitrin was 0.0141-0.6613 mg/mL. Conclusion:The yield of total flavonoids is different with different methods,and the extraction effects of rutin,quercitrin and hyperin are also different. Compared with the four methods used in the experiment,the ethanolic extraction is the most ideal method for the extraction of total flavonoids,rutin and hyperin,while the best method for extracting quercetin is the method of water extraction.
