急进高原对大鼠小肠组织多巴胺2受体影响及生白术的干预研究

摘要
[目的]观察急进高海拔环境Wistar大鼠胃肠道动力的变化,研究其发生机制及生白术的干预作用。[方法]70只Wistar大鼠随机分为7组,对照组(CG组),3500m海拔模型组(3500 MS组),5000m海拔模型组(5000MS组)、3500m生白术大剂量防治组(3500SBZD组),35000m生白术小剂量防治组(3500SBZX组),5000m生白术大剂量防治组(5000SBZD组),5000m生白术小剂量防治组(5000SBZX组),每组10只。除CG组,其余各组依据3500m和5000m海拔要求分别在低压氧舱处理3d,CG组、3500MS组和5000MS组给予生理盐水灌胃,3500SBZD组、3500SBZX组、5000SBZD组和5000SBZX组分别给予不同浓度(540g/L,270g/L)的生白术煎制溶液灌胃,2次/d,2ml/次。3d后给予2ml墨汁灌胃,30min后处死,测量小肠墨汁推进率,检测小肠组织多巴胺2受体(DR2)水平。[结果]与CG组比较,3500MS组小肠推进率降低,DR2增加;5000MS组小肠推进率加快,DR2减少;3500SBZD组和3500SBZX组小肠推进率上升、DR2下降;5000SBZD组和5000SBZX组小肠推进率下降、DR2上升。但3500SBZX组和5000SBZX组作用不明显(P>0.05)。[结论]高海拔环境导致胃肠运动功能紊乱,与DR2有关,生白术可有效调控肠道运动功能,且具有量效关系。
        [Objective]To observe the effect of raw atractylodes on the small intestine tissue and small intestine propulsion rate of rats,discuss the mechanism of the gastrointestinal motility dysfunction and effects of raw atractylodes in the high altitude environment.[Methods]Set of the model of high altitude environment by hypobaric oxygen chamber for 3days.70 Wistar rats were randomly divided into 7groups:the control group(Lanzhou elevation,1430 m,CG group),the model set group(3500m,MS1 and 5000 m,MS2),the big and small raw atractylodes group(3500m,SBZD1 and SBZX1;5000m,SBZD2 and SBZDX2).SBZ group were lavaged 2 mL with raw atractylodes.The others were lavaged 2 mL with normal saline(NS).All of them were lavaged 2mL with ink after 3dand put to death after 30 min.We measured the small intestinal propulsion rates of ink and DR2 by Immunohisto-chemical method.[Results]Compared with the CG,the small intestinal propulsion rates was reduced and the positive area of DR2 was increased in the MS group at 3500 m,but them were opposite at 5000 m.Raw atractylodes could increase the small intestinepropulsion rate and drop the positive area of DR2 at 3500m,decrease the small intestine propulsion rate and increase the positive area of DR2 at 5000m,but the effect of the small raw atractylodes is not obvious(P> 0.05).[Conclusion]The condition of high altitude could induce gastrointestinal motor function disorder.It may be closely associated with the altitude and DR2.Raw atractylodes could effectively adjust the disorder and repair injury.They had dose-effect relationship.
