摘要
目的探讨奥拉西坦对血管性痴呆大鼠认知功能及脑组织N-myc表达的影响。方法将Wistar雄性大鼠分为假手术组10只、模型组10只和奥拉西坦治疗组10只,假手术组不进行模型建立操作,奥拉西坦治疗组腹腔注射10μg/kg奥拉西坦,模型组腹腔注射等量生理盐水,观察与检测大鼠认知功能及脑组织N-myc表达情况。结果模型组与奥拉西坦治疗组均成功建立了血管性痴呆大鼠模型,水迷宫实验显示奥拉西坦治疗组第3天、第5天逃避潜伏期明显低于模型组,差异有统计学意义(P<0.05)。奥拉西坦治疗组与模型组、假手术组的避暗实验潜伏期、错误次数比较差异均有统计学意义(P<0.05)。奥拉西坦治疗组大脑海马组织N-myc表达的吸光度明显高于模型组与假手术组,差异均有统计学意义(P<0.05)。结论奥拉西坦通过上调N-myc蛋白表达,可能对血管性痴呆大鼠海马神经元保护起着至关重要的作用,可作为治疗血管性痴呆的一个新途径。
        Objective To investigate the effects of oxiracetam on brain tissue N-myc expression and nerve regeneration in the vascular dementia rats.Methods Male wistar rats were divided into sham operation group,model group and oxiracetam treatment group with 10 rats in each groups.The sham operation group was not performed model build operations,and the oxiracetam treatment group received oxiracetam by intraperitoneal injection of 10 μg/kg,the model group received intraperitoneal injection of saline.The rat brain N-myc expression was detected and the nerve regeneration organization was observed.Results Vascular dementia rat model were all successfully established in model group and the oxiracetam treatment group,the water maze test showed that the escape latency of the oxiracetam treatment group on day 3 and day 5 was obviously lower than that of the model group,and the result was statistically significant(P<0.05).There were statistically significant difference between oxiracetam treatment group and sham operation group,model group with respect to latency and error frequency(P<0.05).The absorbance of N-myc expression in hippocampus brain in the oxiracetam treatment group was significantly higher than that of model group and sham operation group(P<0.05).Conclusion Oxiracetam may play a crucial role in protection of hippocampal neurons in rats with vascular dementia through upregulation of N-myc protein expression,and may be a new way for the treatment of vascular dementia.
