摘要
目的探讨P-选择素糖蛋白配体1(P-selectin glycoprotein ligand 1,PSGL-1)和CD44在慢性阻塞性肺疾病(chronic obstructive pulmonary disease,COPD)患者外周血中性粒细胞中的表达,以及香烟烟雾提取物对健康志愿者外周血中性粒细胞PSGL-1和CD44表达的影响。方法收集华中科技大学同济医学院附属同济医院2015年9月至2015年12月呼吸与危重症医学科收治的26例COPD患者(COPD组),以及24例无吸烟史的健康志愿者作为正常对照组(NC组),流式细胞术检测COPD患者和健康志愿者外周血中性粒细胞表面PSGL-1和CD44的表达。同时分离6例健康无吸烟史的志愿者外周血标本中的中性粒细胞,用不同浓度(0%、5%、10%、20%)香烟烟雾提取物(cigarette smoke extract,CSE)刺激,流式细胞术检测中性粒细胞表面PSGL-1和CD44的表达。结果 COPD组患者外周血中性粒细胞表面PSGL-1、CD44的表达较NC组明显增强(均P<0.05),CSE刺激的正常人外周血中性粒细胞表面PSGL-1、CD44表达明显增强(均P<0.05),且呈浓度依赖性。结论 COPD患者外周血中性粒细胞表面PSGL-1和CD44的表达较正常对照组增强。香烟烟雾提取物可能通过促进外周血中性粒细胞表面PSGL-1和CD44的表达,从而启动中性粒细胞的粘附级联反应,促进气道中性粒细胞炎症反应。
        Objective To investigate the expression of P-selectin glycoprotein ligand 1(PSGL-1) and CD44 on neutrophils in chronic obstructive pulmonary disease(COPD)patients and the effect of cigarette smoke extract(CSE)on their expression.Methods A total of 26 COPD patients hospitalized in the Department of Respiratory Diseases and Critical Care Medicine in Tongji Hospital from September 2015 to December 2015,and 24 healthy controls were enrolled in this study.The expression of PSGL-1 and CD44 on neutrophils in COPD patients and healthy controls were detected by flow cytometry.In addition,peripheral neutrophils were separated from 6healthy non-smokers volunteers and stimulated with cigarette smoke extract(CSE)at various concentrations(0%,5%,10% and 20%).The expression of PSGL-1 and CD44 on neutrophils was detected by flow cytometry.Results The expression of PSGL-1 and CD44 on neutrophils significantly increased in COPD patients in comparison to healthy controls(both P<0.05).The expression of PSGL-1and CD44 on neutrophils significantly increased with CSE stimulation(both P<0.05),and it was concentration-dependent.Conclusion The expression levels of PSGL-1 and CD44 are significantly increased in COPD patients.CSE increases their expression on neutrophils in human peripheral blood,thus may initiate adhesion cascade reaction of neutrophils,and promotes the neutrophil recruitment in the airway of COPD patients.
