摘要
目的研究Claudin-1、2、3、4在表皮肿瘤特别是在SCC的发生、发展中所起的作用,为从分子水平揭示表皮肿瘤特别是SCC的病因、发病机制、诊断和靶向治疗提供新思路。方法采用免疫组化方法检测正常表皮、脂溢性角化病(SK)、日光性角化病(AK)、鲍温病(BD)和皮肤鳞状细胞癌(SCC)组织中Claudin-1、2、3、4的表达水平。结果Claudin-1在正常表皮和SK中的表达阳性率明显高于AK、BD和SCC,在AK中的表达阳性率明显高于BD和SCC,差异均有统计学意义(P <0.05);Claudin-2在正常表皮、SK、AK、BD和SCC中的表达差异均无统计学意义(P>0.05);Claudin-3、4在正常表皮和SK中的表达阳性率明显低于AK、BD和SCC,在AK中的表达阳性率明显低于BD和SCC,差异有统计学意义(P<0.05)。结论 Claudin-1、3、4的异常表达可能与表皮鳞状上皮的恶性转化相关;联合检测Claudin-1、3、4的表达可能可作为皮肤原位癌及皮肤鳞状细胞癌早期诊断的指标。
Objective To research the role of Claudin-1,2,3,4 in the occurrence and development of skin tumors,especially SCC,and to reveal skin tumors,especially the etiology,pathogenesis of SCC and provide new ideas for diagnosis and targeted therapies.Methods The expression levels of Claudin-1,2,3,4 were investigated in the normal epidermis,seborrheic keratosis(SK),actinic keratosis(AK),Bowens disease(BD)and squamocellular carcinoma of the skin(SCC)by immunohistochemistry.Results The positive expression rate of Claudin-1 in the normal epidermis and SK was significantly higher than that of AK,BD and SCC,and the positive expression rate in AK was significantly higher than that of BD and SCC,with statistically significant differences(P<0.05).The expression differences of Claudin-2 in normal epidermis,SK,AK,BD and SCC were not statistically significant(P>0.05).The positive expression rate of Claudin-3,4 in the normal epidermis and SK was significantly lower than that of AK,BD and SCC,and the positive expression rate in AK was significantly lower than that of BD and SCC,with statistically significant differences(P<0.05).Conclusion Abnormal expression of Claudin-1,3,4 may be associated with malignant transformation of epidermal squamous epithelium.The expression of Claudin-1,3 and Claudin-4 can be used as the indicators for early diagnosis of skin carcinoma in situ and skin squamous cell carcinoma.
