牛磺酸对MDCC-MSB1细胞端粒酶表达水平及其活性的影响

摘要
为探讨牛磺酸对鸡MDCC-MSB1肿瘤细胞系增殖的影响及其可能的作用机理,研究在牛磺酸应用适宜浓度范围内,使用添加0%、0.5%、1.0%、2.0%牛磺酸的RPMI1640培养基,培养MDCCMSB1细胞系,探讨牛磺酸对鸡MD肿瘤细胞chTERT的表达及其端粒酶活性的影响。结果显示,与对照组相比,各牛磺酸组MD肿瘤细胞chTERT转录水平和端粒酶活性均降低,其中1%、2%牛磺酸浓度组的chTERT转录水平和端粒酶活性显著低于对照组(P<0.05)。牛磺酸对chTERT转录水平及端粒酶活性的下调,可能抑制肿瘤细胞的增殖。
        In order to explore effect of taurine on proliferation of MDCC-MSB1 cell and its possible mechanism,In proper concentration range of taurine, the MDCC-MSB1 cell were cultured in RPMI1640 medium with different concentration taurine which is respectively 0%,0.5%,1.0% and 2.0%. Telomerase activity and expression of and chTERT subunit were tested by semi-quantitative reverse transcriptase-polymerase chain reaction(RT-PCR) and TRAP techniques for its effect on the tumor cells.The results showed that telomerase activity and chTERT expression in mRNA level of taurine group cells are down regulated compared with the control group. Telomerase activity and the chTERT expression of 1%, 2% taurine groups are reduced significantly compared with the control group(P< 0.05). Taurine might inhibit proliferation of tumor cell by down regulating telomerase activity and chTERT expression in mRNA level.
