PRRS virus replicates efficiently in splenic CD163+ macrophages. Splenic CD163+ macrophages express Siglec-3 and Siglec-5 but very low levels of CD169. Although PRRS virus binds to Siglec-3, CD169 is the dominant receptor for binding to splenic CD163+ macrophages.
