Résumé
La production d’anticorps monoclonaux est une technique qui a plus de 30 ans et qui continue de subir des améliorations. Les premiers anticorps produits l’ont été à visée diagnostique, initialement appliqués à des techniques d’immunoblotting puis d’immunofluorescence et enfin d’immunohistochimie. On est progressivement passé d’anticorps polyclonaux peu performants (en particulier sur coupe de tissu) à des anticorps monoclonaux murins. En parallèle de leur utilisation diagnostique, les anticorps ont été utilisés pour des applications thérapeutiques (cancer ou maladies inflammatoires) après humanisation partielle ou totale. Les anticorps monoclonaux restent aujourd’hui les outils de diagnostic tissulaire les plus performants. La possibilité récente d’obtention d’hybridomes de lapin a encore permis d’améliorer la qualité des outils disponibles pour le diagnostic histopathologique.
Summary
Since their invention in the seventies, the techniques for generating monoclonal antibodies in mice are constantly improving. The first antibodies applied for diagnosis purposes were first used for immunoblotting techniques. Later antibodies technologies were transferred on tissues sections with immunofluoresence and immunohistochemistry. It turned out that monoclonal antibodies were much more efficient than polyclonal ones, in particular on tissue sections. Beside their use in diagnostic pathology, monoclonal antibodies have been humanized to use them for treatment of several diseases such as cancers and inflammatory disorders. Monoclonal antibodies remain the best tools for diagnosis in routine histopathology. The recent production of rabbit monoclonal antibodies represents an important breakthrough in the field of immunohistochemistry.
