A method for the assay of HEPES was developed and validated. The influence of temperature, solvent strength, pH and ion concentration on the retention behavior was investigated. Four different retention models were applied to the retention data. A degradant forming under oxidative stressing condition was identified as N-oxide of HEPES. The final method was applied to the commercially available samples.
