Berberine binds to nucleosomes with higher affinity than DNA. The interaction between berberine and nucleosomes is positive cooperative and spontaneous. Berberine quenches with globular core domain of histone H1 and nucleosomes chromophores reducing fluorescence intensity. Histone H1 is introduced as a new binding site for berberine in chromatin structure.
