The PSI/PSII chlorophyll (Chl) ratio is measured in leafs with lifetime microscopy. The PSI/PSII Chl ratio is different for the top and bottom of the leaf. Far red-light acclimation reduces the PSI/PSII Chl ratio by a factor of three. The PSI/PSII Chl ratio was visualized within grana and stroma inside the leaf. The method can be used to follow dynamic processes in chloroplasts.
