293: Anti-inflammatory effect of kopetdaghins A, C and E from Dorema Kopetdaghense

Nitric oxide (NO) is synthesized in large quantities by activated inflammatory macrophages and has been demonstrated to be involved in the pathogenesis of inflammatory conditions. Dorema kopetdaghense is endemic to Iran and it has been used in traditional medicine from the ancient time. The present study investigated the potential anti-inflammatory activity of new Dorema kopetdaghense derivatives. New kopetdaghins A, C and E were isolated and their non-toxic concentrations were evaluated for their anti-inflammatory activity. Lipopolysaccharide (LPS)-stimulated J774A.1 macrophages were treated with kopetdaghins and the amount of nitric oxide (NO) production and the expression of inducible nitric oxide synthase (iNOS) were evaluated. Kopetdaghins did not reduce the viability of macrophages. While, they significantly inhibited the enhanced production of NO and the expression of iNOS induced by LPS in a concentration-dependent manner in J774A.1 macrophages. This experiment for the first time revealed the remarkable anti-inflammatory activity of new isolated kopetdaghins from Dorema kopetdaghense.
