Acute pain, short term and long term effects of αO-conotoxin GeXIVA isomers in CCI model were evaluated systematically.

•

GeXIVA isomers produced significantly anti-allodynia in CCI rats without tolerance.

•

Antinociceptive effect of GeXIVA[1,2] in CCI rats was slightly better than GeXIVA[1,4].

•

The pain reliever effect of GeXIVA[1,2] was persisted for two weeks after the cessation of treatment.

• GeXIVA isomers showed no analgesic effect in the tail flick model and have no effect on motor activity in normal rats.
