•: Single molecules of calmodulin (CaM) labeled with Alexa Fluor 488 were detected in live HEK cells.
•: Locations were resolved to < 100 nm to generated super-resolved images of presumed signaling complexes with bound CaM.
•: Single CaM molecules displayed a wide range of mobilities, indicating interactions with cellular components.
•: In cells at high Ca²⁺ CaM was commonly restricted to regions of < 1 μm; at low Ca²⁺ more CaM molecules diffused normally.
