•: The toxicity of MEQ and its main metabolites in vitro in HepG2 cells and in vivo in mouse hepatocytes were investigated.
•: MEQ could induce cell cycle arrest to the G2/M phase and cause dose-dependent DNA damage in HepG2 cells in vitro and in murine hepatocytes in vivo.
•: Serious DNA damage in HepG2 cells induced by the metabolites was not observed.
