Tumor suppressor RYBP harbors three nuclear localization signals and its cytoplasm-located mutant exerts more potent anti-cancer activities than corresponding wild type

RYBP contains three monopartite nuclear localization signals (NLSs) at its N-terminus. A cytoplasm-located mutant of RYBP (RYBPmut) was generated by replacing four basic amino acid residues in these NLSs. RYBPmut showed a more potent ability to bind to caspase 8, and to activate p53 than its wild type in human tumor cells. RYBPmut demonstrates more potent anti-tumor activities than its wild type, in both p53-dependent and independent manners.
