Cbb3-CcOs isolated from CcoQ-deletion mutants show dramatic reduced activity. Subunit CcoQ was removed from the mature wild-type Cbb3-2 complex. Once correctly assembled, Cbb3-CcOs are fully active even in the absence of CcoQ. We propose that CcoQ functions as a chaperone involved in the assembly of Cbb3-CcOs.
