•: We isolated new acrolein toxicity-decreasing Neuro2a (Neuro2a-ATD2) cells.
•: In Neuro2a-ATD2 cells, the IC₅₀ of acrolein increased from 4.2 to 6.8 μM.
•: The mechanism to decrease the toxicity was due to the decrease in AcPAO and SMO.
•: In these cells, the levels of FosB and C/EBPβ transcription factors were reduced.
•: The results indicate that acrolein is mainly produced from polyamines by PAO.
