•: MC-LR was accumulated more in intestine of Procambarus clarkii than other organs and eliminated more quickly in gill than other organs with freshwater depuration.
•: Antioxidant enzymes and biotransformation enzyme played key roles in scavenging MC-LR toxicity.
•: Heat shock protein may be an important cellular defense to decrease oxidative damage induced by MC-LR in crustaceans.
