摘要
利用SCoT分子标记对44份籽用南瓜品种进行遗传多样性分析。从62个引物中筛选出24个有多态性的引物来对44份籽用南瓜品种进行PCR扩增,得到了228个位点,其中具有多态性的位点有168个,占总位点数的73.68%。应用POPGENE 32分析得到,44个籽用南瓜品种的平均Nei’s基因多样性系数为0.3055,平均Shannon’s信息指数是0.4425。利用NTSYS Version2.1软件分析得,所有籽用南瓜品种的相似系数在0.522~0.934之间;进行非加权组平均法(UPGMA)聚类分析得出树状图,将44份籽用南瓜品种完全的区分开来,并在相似系数0.724处,分为8类。从得到的一系列遗传相关系数可说明,SCoT分子标记技术可以很好地应用于籽用南瓜材料的遗传多样性分析。
        SCoT markers were used to study genetic diversity and relationship of 44 seed-used pumpkin accessions. Twenty-four primers were screened from 62 SCoT primers and then used to amplify the genomic DNAs of all the accessions. Totally 228 SCoT loci were produced, including 168 polymorphic loci, with a polymorphism rate of 73.68% obtained by POPGENE 32 software. The average Nei's gene diversity(H) and Shannon's information index(I) were 0.3055 and 0.4425, respectively. The Jaccard genetic similarity coefficients among the accessions were calculated with NTSYS Version 2.1 software and the values were between 0.522 and 0.934. UPGMA cluster analysis was used to draw the dendrogram, and the 44 seed-used pumpkin accessions were completely separated from each other. All test materials were divided into 8 groups at the similarity coefficient of 0.724. The results obtained from a series of genetic correlation coefficients showed that the SCoT molecular marker technique can be used to analyze the genetic diversity of seed-used pumpkin.
