摘要
目的:探讨妊娠激活醛固酮促进肾小管上皮细胞增殖的机制及益肾健脾泻浊中药的保护作用。方法:雌性大鼠按随机数字表法分为假手术组、假手术+妊娠组、UUO组、UUO+妊娠组、UUO+妊娠+螺内酯组、UUO+妊娠+中药组,UUO各组采用结扎单侧输尿管(UUO)的方法复制慢性肾间质纤维化模型,2治疗组分别给予螺内酯和益肾健脾泻浊中药治疗。8周后妊娠各组按动情周期与雄鼠合笼,妊娠第19天处死母鼠,采用放射免疫、免疫组化、Western blot法检测醛固酮、SGK-1、NF-κB、PCNA的表达。结果:益肾健脾泻浊中药可减轻妊娠大鼠肾脏病理损伤,降低血清醛固酮含量,下调SGK-1、NF-κB、PCNA的蛋白表达。结论:益肾健脾泻浊中药可抑制妊娠激活醛固酮诱导的细胞增殖,延缓肾间质纤维化进程。
Objective:To explore the mechanism of pregnancy activated aldosterone stimulated renal tubular epithelial cell proliferation and the protective effect of Yishen Jianpi Xiezhuo recipe.Methods:Female Wistar rats were randomly divided into Sham group,Sham + pregnancy group,UUO group,UUO + pregnancy group,UUO + pregnancy +Spironolactone group,UUO + pregnancy + Chinese medicine group.All of the UUO groups rats reproduce the renal interstitial fibrosis model by unilateral ureteral ligation(UUO).The two treatment groups were given Spironolactone and Yishen Jianpi Xiezhuo recipe respectively.After 8 weeks,the rats in each group of pregnancy were caged with male rats according to the estrus cycle.On the 19 th day of pregnancy,the pregnancy is terminated.The Serum and glucocorticoid induced kinase-1(SGK-1),Nuclear factor-kappa B(NF-κB) and Proliferating Cell Nuclear Antigen(PCNA) by radioimmunoassay,immunohistochemistry and Western blot.Results:Yishen Jianpi Xiezhuo Recipe can reduce renal pathological damage,levels of serum aldosterone,and reduce the expression of PCNA,SGK-1 and PCNA.Conclusion:Yishen Jianpi Xiezhuo Recipe could inhibit the cell proliferation induced by aldosterone in pregnancy rats,delay the progression of renal interstitial fibrosis.
