摘要
目的:建立指纹图谱与一测多评(QAMS)结合的考察模式,验证该方法在中药质量评价中的准确性和可行性。方法:以鹿茸为研究对象,建立鹿茸核酸类成分的HPLC指纹图谱,以次黄嘌呤为参照物计算该成分与尿嘧啶、腺嘌呤、尿苷、肌苷、尿苷的相对校正因子,并计算含量,实现QAMS。同时对QAMS的计算值与外标法实测值进行比较,以确认QAMS法的可行性和科学性。结果:建立了鹿茸核酸类成分HPLC特征指纹图谱共有模式,标定了12个共有峰,指认了其中6个共有峰,15批鹿茸的相似度均大于0.95。15批鹿茸样品中6个主要成分的计算值与实测值无显著差异。结论:QAMS结合指纹图谱的质量控制模式在鹿茸中得到验证,该方法快速准确灵敏,可为鹿茸定量测定和指纹图谱质量控制提供参考。
        OBJECTIVE To establish a new method for the quality evaluation of Velvet antler by fingerprint and quantitative analysis of multi-components by single marker method(QAMS) and validate its accuracy and feasibility for the application in preparation. METHODS The HPLC fingerprint of antler nucleic acid was established by taking pilose antler as the research object. The relative correction factors of the components with uracil, adenine, uridine, inosine and uridine were calculated with hypoxanthine as the reference substance. Meanwhile, the calculated value of QAMS is compared with the measured value of external standard method to confirm the feasibility and scientificity of QAMS method. RESULTS The common pattern of HPLC fingerprint of Velvet antler nucleic acid was established. 12 common peaks were identified and 6 common peaks were identified. The similarity of 15 batches of Velvet antler was more than 0.95. There was no significant difference between the calculated and measured values of the six main components in 15 batches of velvet antler samples. CONCLUSION The method of fingerprint combined with QAMS has been verified in Velvet, which could be used as a reference for the quality control of multiple components determination and fingerprint chromatography of Velvet antler in future.
