台盼蓝、次甲基蓝及健那绿在猪精子活率测定中的比较研究

摘要
本试验采用不同浓度的台盼蓝、次甲基蓝、健那绿对猪精子进行染色,以测定猪的精子活率。结果表明,用三种染液染色后,死精子和活精子各自表现出不同的染色特征。台盼蓝和次甲基蓝的染色效果为:活精子透明不着色,死精子着蓝色;健那绿的染色效果为:活精子尾部中段着蓝绿色、细胞质呈无色,死精子着绿色。在不同染色条件下比较得出:0.4%台盼蓝染色15min的染色效果最好。用次甲基蓝、健那绿试剂染色测得的精子活率明显低于实际精子活率,因此次甲基蓝、健那绿不可用于猪精子的活体染色。
        In this study,the sperm viability of pigs was determined by different concentrations of Trypan blue, Methylene blue and Janus green. The results showed that after staining pig sperm with three kinds of dyes, dead sperm and live sperm showed different staining characteristics. The dyeing effect of Trypan blue and Methylene blue was:live sperm was not stained,dead sperm was blue;the dyeing effect of Janus green was:live sperm tail was blue and green,the cytoplasm was colorless,and dead sperm was green. Through the comparison of different staining time and different dyeing concentration, 0.4% Trypan blue staining for 15 min had the best dyeing effect. The sperm viability of dyeing Methylene blue and Janus green was lower than that of the actual sperm viability. So Methylene blue,Janus green can′t be used for living staining of pig sperm.
