豁痰化瘀利水方对急性硬膜下血肿大鼠脑组织MCP-1、NF-κB的影响

摘要
目的探究豁痰化瘀利水方对急性硬膜下血肿(ASDH)大鼠脑组织单核细胞趋化因子-1(MCP-1)、核因子-κB(NF-κB)的影响。方法将成年SD大鼠随机分为模型组(ASDH组)、假手术组(Shame组)、天丹通络胶囊组(阳性对照组)及豁痰化瘀利水方组(观察组),其中观察组依据剂量可分为低剂量、中剂量、高剂量组; ASDH造模成功后给予各组相应治疗处理,观察脑组织硬膜下血肿面积,并检测急性硬膜下脑组织中MCP-1、NF-κB阳性表达情况、mRNA表达水平及蛋白水平。结果治疗处理后,与ASDH模型组比较,其他组血肿面积、MCP-1、NF-κB阳性细胞表达比例、mRNA相对表达水平及蛋白水平均显著降低(P<0.01);与阳性对照组比较,中剂量、高剂量组血肿面积、MCP-1、NF-κB阳性细胞表达比例、mRNA相对表达水平及蛋白水平均显著降低(P<0.01),而低剂量组血肿面积、MCP-1、NF-κB相对表达水平均显著升高(P<0.01);与中剂量比较,高剂量组血肿面积、MCP-1、NF-κB阳性细胞表达比例、mRNA相对表达水平及蛋白水平均显著降低(P<0.01)。结论豁痰化瘀利水方能够有效减少急性硬膜下血肿面积,降低脑组织中MCP-1、NF-κB的表达,其效果与豁痰化瘀利水方剂量关系密切。
        Objective To explore the influence of removing phlegm,removing stasis,inducing diuresis decoction on MCP-1 and NF-κB in brain tissue of rats with acute subdural hematoma(ASDH).Methods The adult SD rats were randomly divided into the model group(ASDH group),the sham operation group(Shame group),the Tiandan Tongluo capsule group(positive control group),and the removing phlegm,removing stasis,inducing diuresis decoction group(observation group).The observation group were divided into low dose,medium dose,high dose groups according to basis dose.After ASDH model was built successfully,all the groups were given the corresponding treatment,and hematoma area size in brain subdural tissue was observed,and MCP-1 and NF-κB positive expressions,m RNA expression and protein level in acute subdural brain tissue were detected.Results After treatment,compared with those in the ASDH model group,hematoma area,MCP-1,NF-κB positive cells expression ratio,m RNA relative expression level and and protein level significantly decreased in the other groups(P<0.01).Compared with those in the positive control group,hematoma area,MCP-1,NF-κB positive cells expression ratio,m RNA relative expression level and and protein level significantly decreased in the medium dose,high dose groups of the observation group(P<0.01),and hematoma area,MCP-1,NF-κB relative expression level significantly increased in the low dose group(P<0.01).Compared with those in the medium dose group,MCP-1,NF-κB positive cells expression ratio,m RNA relative expression level and and protein level significantly decreased(P<0.01).Conclusion Removing phlegm,removing stasis,inducing diuresis decoction can effectively reduce the area of acute subdural hematoma,reduce the expression of MCP-1 and NF-κB in brain tissue.The effect is closely related to the dosage of removing phlegm,removing stasis,inducing diuresis decoction.
