摘要
为研究油菜、虞美人、柳树等3种蜂花粉的总黄酮提取物(total flavonoids of bee pollen, TFBP)的自由基清除能力,比较不同蜂花粉之间的抗氧化能力,并探究总黄酮含量与自由基清除能力之间的关系,进而筛选出具有理想抗氧化活性的蜂花粉品种,本研究采用超声醇提取大孔树脂纯化的方法提取3种蜂花粉的TFBP,同时采用分光光度法测定其中总黄酮的含量。以槲皮素作为阳性对照品,通过测定梯度稀释的各样品对DPPH,·OH,·O_2~-及ABTS+等4种自由基的清除率,评价各种蜂花粉TFBP的自由基清除能力,并对蜂花粉的总黄酮含量及各种自由基的清除活性进行相关性分析。结果表明:3种蜂花粉TFBP对DPPH自由基的清除能力排序为柳树>油菜>虞美人,对·OH自由基的清除能力排序为油菜>柳树>虞美人,对·O_2~-自由基的清除能力排序为油菜>柳树>虞美人,对ABTS+自由基的清除能力排序为油菜>虞美人>柳树。相关性分析结果显示,蜂花粉总黄酮含量与·OH、·O_2~-及ABTS+自由基的清除能力均有很强的相关性,但与DPPH自由基清除能力的相关性较弱。在3种蜂花粉中,油菜蜂花粉TFBP表现出较强的自由基清除活性,具有良好的抗氧化潜力,可作为开发效果理想的天然抗氧化剂的备选材料。另外,相关性分析指出,蜂花粉的自由基清除能力不仅仅与其总黄酮含量有关。
        In this paper, the total flavonoids of bee pollen(TFBP) of three species(rape, coquelicot, and willow) were researched.By evaluating their free radical scavenging capacities, the differences in antioxidant activities among different bee pollens were compared, and the correlation between total flavonoids content and free radical scavenging capacities was explored. Furthermore,the bee pollen varieties with ideal antioxidant activity were screened to provide a reference for the development of natural antioxidants with excellent performance. Experimental methods: Ultrasound-assisted alcohol extraction and macroporous resin purification were used to extract and purify the TFBP from 3 kinds of bee pollens, and the content of total flavonoids was determined by spectrophotometry. Quercetin as a positive control, the free radical scavenging capacities of various TFBPs were evaluated by measuring the scavenging rates of four kinds of free radicals such as DPPH, ·OH, ·O_2~- and ABTS+. Then,correlation analysis between total flavonoids content and free radical scavenging capacities of bee pollen were performed. The results showed that the DPPH radical scavenging capacities of 3 TFBPs were willow>rape>coquelicot. The ·OH radical scavenging capacities of 3 kinds of TFBPs were rape>willow > coquelicot. The ·O_2~- radical scavenging capacities of 3 kinds of TFBPs were rape>willow>coquelicot. The ABTS+radical scavenging capacities of 3 kinds of TFBPs were rape >coquelicot >willow. Correlation analysis showed that the total flavonoids content had a strong correlation with the scavenging capacities of ·OH, ·O_2~- , and ABTS+radicals, but a weak correlation with DPPH radical. Conclusion: Among the 3 species of bee pollen, rape TFBP showed a stronger free radical scavenging activity than the other 2 bee pollens, and had an excellent antioxidant potential, which could be used as an ideal alternative material for developing natural antioxidants. In addition, the correlation analysis pointed out that the free radical scavenging activity of bee pollen was not only related to the total flavonoids content.
