摘要
建立洋葱皮醇提物(OPE)总多酚含量测定方法并评价OPE不同部位总多酚含量及其降脂和体外抗氧化活性。结果表明,OPE总多酚含量测定的最佳显色条件为:显色时间80 min,显色温度85℃,10%碳酸钠溶液用量0.9 mL,福林试剂用量1.2 mL;没食子酸在(1.2~5.0)mg/L范围内呈良好的线性关系,R~2为0.9977,平均回收率为100.489%(n=6),计算得洋葱皮总多酚含量为10.830mg GAE/g DW。系统溶剂萃取法后得到正丁醇萃取部位(OPB)、水部位(OPW)和二氯甲烷萃取部位(OPC)的总多酚含量分别是6.526、3.629、0.345 mg GAE/g DW;体外HepG2降脂活性筛选实验中,洋葱皮醇提物各部位均有不同程度的降脂活性(P<0.05);洋葱皮醇提物各部位还具有不同程度的抗氧化活性,其中OPB具有最强的抗氧化活性,比阳性药芦丁的抗氧化活性强。由此可见,OPB具有最高的总多酚含量,最强的抗氧化活性,且具有一定的降脂活性,具有进一步开发利用的价值。
        The determination method of the total polyphenol content from onion peel extract(OPE) was established. The lipid-lowering and antioxidant activities of the different parts of OPE were evaluated.As a result, the best colour-developing conditions of determinating the total polyphenol content were as follows: 80 min(chromogenic time), 85 ℃(chromogenic temperature), 1.2 mL(volume of Folinciocalteure agent), 0.9 mL(volume of 10% Na_2CO_3 solution). Gallic acid showed a good linear relationship in the range of(1.2~5.0)mg/L(R~2=0.9977). The average recovery rate was 100.489%(n=6). The total polyphenol content was 10.83 mg GAE/g DW. After systematically extracting with different solvents, the total polyphenol content of n-butanol part(OPB), water part(OPW) and dichloromethane part(OPC) was 6.526, 3.629, 0.345 mg GAE/g DW, respectively. In the screening model of HepG2 lipid-lowering activity in vitro, the various parts of onion peel extracts displayed certain lipid-lowering activity; the onion peel extracts of each parts also exhibited different degrees of antioxidant activity. OPB pocessed the strongest antioxidant activity, which is stronger than that of the positive rutin. Thus, OPB showed the highest total polyphenol content and the strongest antioxidant activity, and processed a certain lipid-lowering activity,which was deserved to develop and utilize further.
