摘要
目的为了探讨辣椒碱(capsaicin)抗小鼠抑郁样作用及其可能的作用机制。方法成年雄性小鼠每天1次灌胃辣椒碱,连续7 d。第7天给药后1 h进行强迫游泳实验(forced swimming test,FST)、悬尾实验(tail suspension test,TST)或旷场实验(open field test,OFT)。FST行为学实验结束后断头迅速取双侧海马、皮质前额叶。比色法和酶联免疫吸附法(ELISA)分别测定海马(hippocampus)及皮质前额叶(prefrontal cortex,PFC)中单胺氧化酶(monoamine oxidase,MAO)活性和5-羟色胺(5-hydroxytryptamine,5-HT)含量。结果辣椒碱显著缩短小鼠在FST和TST中的不动时间,但对OFT中的水平活动时间和活动距离均无明显影响;同时降低FST小鼠的海马和PFC中MAO活性,升高其5-HT含量。结论辣椒碱对小鼠可能有一定的抗抑郁样作用,其机制可能与抑制海马及皮质前额叶MAO活性、增加其5-HT神经递质含量有关。
        Objective To investigate the antidepressant-like effects of capsaicin in mice and its possible mechanisms.Methods Mice were randomized into five groups:three groups were orally treated with different doses of capsaicin,the fourth group was(control group)treated with vehicle and the fifth group received fluoxetine,once a day for 7 consecutive days.One hour after the last administration,the immobility in the forced swimming test(FST)and the tail suspension test(TST),and the locomotion activity in open field test(OFT)were detected,respectively.After the FST behavioral experiment,the bilateral hippocampus and prefrontal cortex of the mice were immediately dissected and stored at-80 ℃ for subsequent determination.The activities of monoamine oxidase(MAO)and the levels of 5-hydroxytryptamine(5-HT)in the hippocampus and prefrontal cortex were respectively determined by the colorimetric method and enzyme linked immunosorbent assay(ELISA).Results As compared with the control group,the immobilities were significantly reduced in the FST and TST.The MAO activities were decreased and 5-HT contents were increased in the hippocampus and prefrontal cortex after capsaicin treatment.Conclusion Capsaicin can relieve depression-like behavior in mice and its mechanism may be related to inhibition of monoamine oxidase activity and regulation of 5-HT levels in the hippocampus and prefrontal cortex.
