摘要
目的制备小型巴马猪椎间盘退行性变模型,观察低强度激光修复术对小型巴马猪退行性变椎间盘水通道蛋白-1表达的影响。方法选取小型巴马猪9只,随机分入对照组(Con组)、造模组(Mod组)及激光治疗组(Las组),每组3只。Con组不做任何处理,观察2个月;Mod组在C形臂X光机引导下,用16 G针刺入猪L2-3、L3-4、L4-5、L5-6椎间盘进行破坏,建模后观察2个月;Las组建模方法与Mod组相同,建模后1个月,对已处理的椎间盘进行低强度激光修复术治疗1个月,再观察1个月。3组巴马猪均接受影像学MR、HE染色、免疫组化、Western Blot等检测,观察椎间盘影像学、组织形态学及水通道蛋白-1表达变化。结果 Con组椎间盘结构完整,Mod组椎间盘结构遭到破坏,Las组椎间盘结构较Mod组显著恢复。与Con组比较,Mod组、Las组的水通道蛋白-1表达水平明显降低,差异均有统计学意义(P<0.05);与Mod组比较,Las组的水通道蛋白-1表达水平明显升高,差异有统计学意义(P<0.05)。结论低强度激光修复术对小型巴马猪退行性变椎间盘具有一定的治疗作用,其机制可能与上调椎间盘组织水通道蛋白-1表达有关。
        Objective To investigate the effect of low intensity laser irradiation on porcine intervertebral disc aquaporin-1(AQP-1) expression by constructing the miniature Bama pig degenerative disc model.Methods Nine miniature Bama pigs were randomly divided into three groups:control group(Con group),model group(Mod group) and laser treatment group(Las group),with 3 pigs in each group.The Con group did nothing and was observed for 2 months.In the Mod group,under the guidance of C-arm X-ray machine,16 G acupuncture was used to Pierce L2-3,L3-4,L4-5 and L5-6 intervertebral discs of the pig for destruction.After modeling,observation was conducted for 2 months.Las group was constructed in the same way as the Mod group.One month after modeling,the treated intervertebral disc was treated with low-intensity laser repair for one month and then observed for another month.MR,HE staining,immunohistochemistry and Western Blot were used to detect the changes of intervertebral disc imaging,histomorphology and AQP-1 expression in all 3 groups of Bama pigs.Results The intervertebral disc structure of Con group was intact,the intervertebral disc structure of Mod group was destroyed,and the intervertebral disc structure of Las group was significantly recovered compared with that of Mod group.Compared with Con group,the expression levels of AQP-1 in Mod group and Las group were significantly reduced,and the differences were statistically significant(P<0.05).Compared with the Mod group,the expression level of AQP-1 in Las group was significantly increased(P<0.05).Conclusion Low intensity laser has a protective effect on degenerated intervertebral disc of Bama miniature pig,and its mechanism is possibly related to the up-regulation of aquaporin expression.
