葛根芩连汤对2型糖尿病大鼠宏观表征及胰高血糖素样肽相关指标影响

摘要
目的:研究葛根芩连汤对2型糖尿病大鼠宏观表征及胰高血糖素样肽水平的影响。方法:选取无特定病原体级SD雄性大鼠40只为研究对象。正常饲养1周后,随机分出10只作为对照组,其余30只为糖尿病研究组,随机分为三组,每组10只大鼠:GGCL1组按照9. 1g/kg体重标准灌胃给葛根岑莲汤药,GGCL2组按照一组按照18. 2g/kg体重标准灌胃给葛根岑莲汤药,模型组灌胃给予等量生理盐水,连续给药6周,观察记录大鼠的体重等一般病理特征及大鼠宏观表征、血糖含量及胰高血糖素样肽水平等指标。结果:模型组大鼠体重降低明显,GGCL1组大鼠体重略有下降,GGCL2组大鼠体重保持稳定; GGCL1、GGCL2两组大鼠给药4周、6周后空腹血糖明显低于模型组,差异有统计学意义(P<0. 05);宏观表征:与模型组大鼠比较,GGCL1组大鼠耳色浅粉、耳血管可见度适中、尾色淡白、大便黑黄,差异有统计学意义(P<0. 05),GGCL2组大鼠活动程度增加、耳色浅粉、耳血管可见度适中、鼻色淡红、舌体适中、爪粉红不润、尾色黄中有白、大便黑黄,差异有统计学意义(P<0. 05); GGCL1、GGCL2组大鼠胰高血糖素样肽水平明显高于模型组大鼠,差异有统计学意义(P<0. 05)。结论:葛根芩连汤能维持2型糖尿病大鼠体重、降低空腹血糖值,增加注射胰岛素后血糖下降速度,提升胰高血糖素样肽水平,对2型糖尿病具有明显的治疗作用。
        Objective: To study the effect of Gegen Qinlian Decoction on macroscopic characterization and glucagon-like peptide for type 2diabetic rats. Methods: Forty male SD rats without specific pathogen were selected as the study subjects. One week after normal feeding,10 rabbits were randomly divided into controlled group and the other 30 rabbits were divided into three groups,10 rats in each group: GGCL1 group was given Gegen Qinlian Decoction according to 9. 1g/kg body weight and GGCL2 group were given Gegen Qinlian Decoction according to 18. 2g/kg body weight,model group was given gavage with normal saline for 6weeks. The general pathological features such as body weight were observed and recorded. Macroscopic characterization of rats,blood glucose levels and glucagon-like peptide levels and other indicators. Results: The body weight of rats in the model group decreased obviously. The body weight of rats in the GGCL1 group decreased slightly. The body weight of rats in the GGCL2 group remained stable. The rats in the GGCL1 and the GGCL2 group were given for 4weeks. After 6weeks,fasting blood glucose was significantly lower than the model group and had statistical significance(P<0. 05). Macroscopical characterization:compared with the model group,the GGCL1 group had mild ear-color powder,moderate visibility of the ear vessels,pale tail color and stool black and yellow,the difference was statistically significant(P<0. 05). In the GGCL2 group,the degree of activity of the rats increased,the ear color was light,the visibility of the ear vessels was moderate,the nose color was pink,the tongue body was moderate,the claw pink was not moist,the tail color yellow was white,the stool was black and yellow,the difference was statistically significant(P<0. 05). The levels of glucagon-like peptide in the GGCL1 and the GGCL2 groups were significantly higher than those in the model group(P < 0. 05). Conclusion: Gegen Qinlian Decoction can maintain the body weight of type 2diabetic rats,reduce fasting blood glucose levels,increase the rate of blood glucose declines after injection of insulin,enhance the level of glucagon-like peptide,has obvious therapeutic effect on type 2diabetes.
