子宫内膜癌病灶内MAP2K4、Fat-1表达变化与癌细胞增殖、侵袭的相关性分析

摘要
目的研究子宫内膜癌病灶内MAP2K4、Fat-1表达变化与癌细胞增殖、侵袭的相关性。方法选择2014年6月至2017年9月进行手术切除的子宫内膜癌患者56例并收集子宫内膜癌病灶及癌旁病灶,选择同期进行手术切除的子宫良性病变患者34例并收集正常子宫内膜。测定组织中MAP2K4、Fat-1及增殖基因、侵袭基因的mRNA表达量。结果子宫内膜癌病灶内MAP2K4、PCNA、CyclinD1、c-erbB2、Vimentin、MMP-9、ADAM19的mRNA表达量显著高于癌旁病灶及正常子宫内膜,Fat-1、pRb2/p130、p27、Caspase-3、E-cadherin、EFEMP2、TIMP1的mRNA表达量显著低于癌旁病灶及正常子宫内膜(P<0.05);Fat-1高表达的子宫内膜癌病灶内PCNA、CyclinD1、c-erbB2的mRNA表达量显著低于Fat-1低表达的子宫内膜癌病灶,pRb2/p130、p27、Caspase-3的mRNA表达量显著高于Fat-1低表达的子宫内膜癌病灶(P<0.05);MAP2K4高表达的子宫内膜癌病灶内Vimentin、MMP-9、ADAM19的mRNA表达量显著高于MAP2K4低表达的子宫内膜癌病灶,E-cadherin、EFEMP2、TIMP1的mRNA表达量显著低于MAP2K4低表达的子宫内膜癌病灶(P<0.05)。结论子宫内膜癌病灶内高表达的MAP2K4和低表达的Fat-1分别与癌细胞侵袭活力增强和增殖活力增强有关。
        Objective To investigate the changes of expression of MAP2 K4 and Fat-1 in endometrial cancer(EC) and their correlation with the proliferation and invasion of cancer cells.Methods The patients with EC who underwent surgical resection in our hospital from June 2014 to September 2017 were enrolled in the study,and the specimens of EC lesions and paracancerous lesions were collected.Meanwhile the specimens of normal endometrium in patients with benign uterine lesions who underwent surgical resection during the same period were collected. The mRNA expression levels of MAP2 K4, Fat-1, proliferative genes and invasive genes in the tissues were measured. Results The mRNA expression levels of MAP2 K4, PCNA, CyclinD1, c-erbB2, Vimentin, MMP-9 and ADAM19 in EC lesions were significantly higher than those in adjacent lesions and normal endometrium,however,the expression levels of of Fat-1, pRb2/p130, p27 and Caspase-3, E-cadherin, EFEMP2 and TIMP1 were significantly lower than those in paracancerous lesions and normal endometrium(P<0.05). The mRNA expressions of PCNA, CyclinD1 and c-erbB2 in EC with high expression of Fat-1 were significantly lower than those with low expression of Fat-1, but the mRNA expressions of pRb2/p130, p27 and Caspase-3 were significantly higher than those with low expression of Fat-1(P<0.05). The mRNA expression levels of Vimentin, MMP-9 and ADAM19 in EC with high expression of MAP2 K4 were significantly higher than those with low expressions of MAP2 K4. The mRNA expression levels of E-cadherin, EFEMP2 and TIMP1 were significantly lower than those with MAP2 K4 low expression(P<0.05).Conclusion The high expression of MAP2 K4 and low expression of Fat-1 in EC lesions are associated with enhancement of invasive activity and proliferative activity of cancer cells.
