摘要
目的探讨长链非编码HOXA11-AS在胃癌及癌旁正常组织中的表达及其在胃癌细胞系中的生物学功能。方法实时荧光定量PCR验证胃癌组织及胃癌细胞系中HOXA11-AS的表达量;核质分离实验及荧光定量PCR检测胃癌细胞胞核与胞质中HOXA11-AS的分布量,明确HOXA11-AS的亚细胞定位;用慢病毒构建HOXA11-AS过表达质粒,筛选稳转株进行细胞功能学实验:细胞增殖、细胞凋亡及细胞周期、细胞迁移能力、细胞侵袭能力。结果 HOXA11-AS在胃癌组织及胃癌细胞的表达量升高(P<0.05);核质分离实验表明,HOXA11-AS在胃癌细胞胞质的分布量高于细胞核(P<0.05);过表达lncRNA HOXA11-AS可促进胃癌细胞的增殖、迁移、侵袭能力(P<0.05),抑制细胞凋亡及阻滞细胞周期。结论 HOXA11-AS在胃癌中高表达,可促进胃癌细胞的肿瘤细胞行为,可作为胃癌治疗的候选分子靶点。
Objective To investigate the expression and biological function of long non-coding RNA( lncRNA)HOXA11-AS in gastric cancer. Methods The expression of HOXA11-AS in gastric cancer and gastric cancer cell lines was determined by real-time PCR. Cytoplasm-nucleus separation and real-time PCR were applied to determine the distribution of HOXA11-AS in cytoplasm and nucleus separated from gastric cancer cell lines. HOXA11-AS expression vector was constructed with lentivirus and transfected into gastric cancer cell lines. Screened for cell lines for stable expression and conducted functional experiments in vitro: cell proliferation,cell apoptosis and cell cycle,cell migration and invasion ability. Results HOXA11-AS in gastric cancer and gastric cancer cells lines were upregulated( P <0. 05). Subcellular fractionation analysis showed that the ratio of HOXA11-AS in the cytoplasm was higher than that in nucleus( P < 0. 05). The upregulated lncRNA HOXA11-AS could promote the proliferation,migration and invasion of HGC-27 cells,and inhibit apoptosis and promote cell cycle progression. Conclusion HOXA11-AS in gastric cancer is upregulated,it can promote the malignant behavior of the tumor cells in gastric cancer,possibly as a candidate molecular target for the treatment of gastric cancer.
