摘要
目的:建立华中枸骨叶药材中6个绿原酸类指标性成分的含量测定方法。方法:采用UPLC法,色谱柱为Thermo Syncromis C18色谱柱(100 mm×3 mm,1.7μm),流动相为0.2%甲酸水溶液(A)-乙腈(B),采用梯度洗脱,流速为0.40 mL·min~(-1),检测波长为327 nm,柱温为40℃。结果:10 min内新绿原酸、绿原酸、隐绿原酸、异绿原酸A、异绿原酸B和异绿原酸C等6个指标性成分分离度良好,且在各自的检测范围内与峰面积积分值呈现良好的线性关系,其相关系数均大于0.9999,加样回收率为97.77%~98.37%。结论:所建立的含量测定方法快速、简便、准确度高且重现性好,适用于华中枸骨叶药材的含量测定与质量控制。
Objective: To establish an UPLC-DAD method for simultaneous determination of 6 chlorogenic acid s in leaves of Ilex centrochinensis. Methods: The separation was achieved on a Thermo Syncromis C18 column(100 mm×3 mm, 1.7 μm) with a gradient elution of 0.2% formic acid(A) and acetonitrile(B) at a flow rate of 0.40 mL·min~(-1). The detection wavelength was 327 nm and the column temperature was 40℃. Results:Satisfactory chromatogram with good separation of 6 phenolic acids including neochlorogenic acid, chlorogenic acid, cryptochlorogenic acid, chlorogenic acid A, chlorogenic acid B and chlorogenic acid C was obtained within10 minutes. All the calibration curves showed good linearity between concentration of each compound and peak area. And the values of r were no less than 0.9999 for all the analytes. The recoveries were 97.77%~98.37%.Conclusion: The developed method is rapid, accurate and reliable, and can be used to determine chlorogenic acids and quality control of leaves of Ilex centrochinensis.
