摘要
目的探讨原钙粘蛋白(PCDH10)在胃癌组织中的表达特点及与患者临床特征、预后的关系。方法分别选取术后胃癌组织标本90例和癌旁组织标本45例,采用蛋白质印迹法、免疫组化法检测两组标本中PCDH10蛋白表达情况,比较不同临床特征的胃癌患者胃癌组织的PCDH10阳性表达率,探讨PCDH10蛋白表达与患者预后的关系。结果胃癌组织中PCDH10阳性表达率为32.22%,明显低于癌旁组织的84.44%,差异有统计学意义(P﹤0.01);胃癌组织中PCDH10蛋白相对表达量明显低于癌旁组织,差异有统计学意义(P﹤0.01);TNM分期为Ⅰ～Ⅱ期、无淋巴结转移的胃癌患者胃癌组织的PCDH10阳性表达率高于TNM分期为Ⅲ期、有淋巴结转移的胃癌患者,差异均有统计学意义(P﹤0.05);PCDH10阳性表达患者的3年生存率为62.1%,高于PCDH10阴性表达患者的36.1%,差异有统计学意义(P﹤0.05)。结论PCDH10在胃癌组织中相对表达量较低,并且与肿瘤的TNM分期、有无淋巴结转移及患者远期预后有关。
Objective To investigate the expression of protocadherin 10(PCDH10) in gastric cancer tissues and its relationship with the clinical characteristics and prognosis of gastric cancer patients.Method A total of 90 cancer tissues specimens collected from gastric cancer patients after surgery,and another 45 specimens of adjacent tissues that met the requirements for postoperative follow-up were selected.Western blot and immunohistochemical assay were used to detect the PCDH10 protein expression in the two groups of specimens,the positive expression rate of PCDH10 protein in patients with different clinical characteristics of gastric cancer,and the relationship between PCDH10 protein expression and patients' prognosis were investigated.Result The positive expression rate of PCDH10 protein in gastric cancer tissues(32.22%) was significantly lower than that in adjacent tissues(84.44%),and the difference was statistically significant(P<0.01).The relative expression of PCDH10 protein in gastric cancer tissues was significantly lower than that in adjacent tissues,with statistically significant difference observed(P<0.01);the positive expression rates of PCDH10 protein in gastric cancer tissues with TNM stage I-II and without lymph node metastasis were significantly higher than those with TNM stage III and lymph node metastasis,indicating statistically significant difference(P<0.05);the 3-year survival rate was 62.1% in patients with PCDH10 positive expression,and was significantly higher than the 36.1% in patients with PCDH10 negative expression,and the difference was of statistical significance(P<0.05).Conclusion The relative expression of PCDH10 is down-regulated in gastric cancer tissues,and is closely related to the TNM stage,presence of lymph node metastasis,and long-term prognosis of patients.
