摘要
目的建立高效液相色谱-蒸发光散射检测法检测保健食品中磷脂酰丝氨酸的含量。方法将样品用氯仿+甲醇(9:1,V:V)提取液溶解定容后过膜,采用高效液相色谱法(正相色谱柱,蒸发光散射检测器)进行检测,流动相为正己烷+异丙醇+水+乙酸+三乙胺(36:55:8:1.5:0.08,V:V:V:V:V),外标法定量。结果在40~1000μg/m L的范围内磷脂酰丝氨酸浓度的自然对数与峰面积的自然对数线性关系良好,相关系数为0.9991,加标回收率为99.00%~100.15%,相对标准偏差为0.3%~0.7%。结论该方法操作简便、准确、重现性好,适用于保健食品中磷脂酰丝氨酸含量的测定。
        Objective To establish a method for determination of phosphatidylserine in health food by high performance liquid chromatography-evaporative light scattering detection. Method The sample was dissolved in extract solution with chloroform/methanol(9:1, V:V) to a fixed volume and then passed through the membrane and detected by high performance liquid chromatography(normal phase chromatographic column, evaporative light scattering detector). The mixture of n-hexane, isopropanol, water, acetic acid and triethylamine(36:55:8:1.5:0.08, V:V:V:V:V) was chosen as the mobile phase and quantified with external standard method. Results The natural logarithm of phosphatidyl serine concentration in the range of 40-1000 μg/m L had a good linearity with the natural logarithm of the peak area, and the correlation coefficient was 0.9991. The recoveries were 99.5%-100.15% and the relative standard deviations were 0.3%-0.7%. Conclusion The method is simple, accurate and reproducible, which is suitable for the determination of phosphatidylserine in health food.
