摘要
目的研究邻苯二甲酸二(2-乙基己基)酯(di-2-ethylhexyl phthalate,DEHP)对GC-2 spd细胞FasL蛋白和mRNA表达的影响。方法将处于对数生长期的GC-2spd细胞,分别暴露于含终浓度为0(溶剂对照)、50、100、200μmol/L DEHP的培养液培养24 h。分别用q RT-PCR及Western blotting检测GC-2 spd细胞FasL mRNA和蛋白的表达。结果 100、200μmol/L DEHP染毒组GC-2 spd细胞FasL mRNA和蛋白的表达水平均高于溶剂对照组,差异有统计学意义(P<0.05);且随着DEHP染毒浓度的升高,GC-2 spd细胞FasL mRNA和蛋白的表达水平呈上升趋势。结论 DEHP可能通过增强生精细胞FasL mRNA及蛋白表达,激活Fas/FasL信号通路,最终导致生精细胞凋亡。
        Objective To study the effects of di-2-ethylhexyl phthalate(DEHP) on protein and mRNA expression of Fas ligand in GC-2 Spd cells. Methods GC-2 spd cells were respectively exposed to the DEHP medium with the final concentrations of0,50,100 and 200 μmol/L for 24 h. The mRNA expression of Fas ligand was measured by real-time PCR,and the protein expression of Fas ligand was tested by Western blotting. Results Compared with the control group,the expression levels of the mRNA and the protein of Fas ligand in the 100 and 200 μmol/L DEHP groups were higher,with significant differences(P <0.05). Conclusion DEHP can induce the apoptosis of GC-2 spd cells through increasing the expression of protein and mRNA of Fas ligand to activate the signal pathway of Fas/FasL.
